The laminin receptor complex (integrin-laminin, Mr = 360,000) is expressed on many cell types and represents a major subgroup of the beta integrin family. Its beta 1 subunit, alpha 6, and gamma 1 chain are essential for laminin attachment. Receptor activity requires both a basal and a laminin-stimulated state. Previous studies have proposed that two major structural components are associated with the basal state, the alpha 6 beta 1 and the alpha 6 gamma 1 molecules. Biochemical analyses of the native receptor complex have identified four proteins, Mr = 115,000 (alpha 6 subunit), Mr = 90,000 (beta 1 subunit), Mr = 65,000 (alpha 6 gamma 1 subunit), and Mr = 42,000 (gamma 1 subunit). The detergent-solubilized receptor appears to be capable of binding laminin in the absence of its associated cytoskeletal proteins, and such receptor activity is insensitive to treatment with the phorbol ester 12-O-tetradecanoyl phorbol-13-acetate. The laminin receptor complex has been purified to apparent homogeneity from the secreted polypeptide-processing cell line PC12W. A similar receptor complex is also detectable in murine fibroblasts and also in murine tissues, including the kidney, lung, skeletal muscle, and liver. In the present study, the physical association of the components of the laminin receptor complex have been examined. The receptor complex was detected in a complex of Mr = 220,000 by immunoprecipitation with the monoclonal antibodies 3E1 and 3C2.
