The nitrogen fixation protein nifH, a key enzyme for dinitrogen (N2) fixation, is tightly controlled at the transcriptional level. The nifH gene in Rhodococcus elongatus CH34, an N2-fixing bacterium, was cloned and the promoter region determined. Deletion of the nifF gene resulted in a 60% decrease in nifH expression, which also impaired the expression of the nifUSV, PFOR, NAP and ferredoxin genes. Deletion of the glnR gene, which is directly or indirectly repressed by NifF, resulted in a 30% decrease in the expression of nifH, nifF, nifUSV, PFOR and NAP. In addition, the nifE gene, which encodes a homologue of ArcB of N2-fixing Gram-positive bacteria, was transcribed under the control of the nifUSV, PFOR and NAP promoters.
