Leaf senescence is characterized by distinctive biochemical and physiological changes. One of the most noticeable changes is the cessation of photosynthesis, leading to degradation of chlorophyll and decreases in the levels of the photosynthetic enzymes. To determine whether OsWOX4 plays a role in regulating photosynthetic activity in rice leaf, we characterized the photosynthetic properties of WT and OsWOX4 knockdown plants. Since OsWOX4 is expressed in the early leaf stage, we produced a pACT1-GVG::OsWOX4:RNAi construct in the Koshiharu background, then treated the seedlings with DEX for 5 days ( Fig 3A ), to knock down OsWOX4 expression. The levels of the photosynthetic enzyme components decreased slightly in the WT. In contrast, these levels were very low in the transgenic plants that had been treated with DEX ( Fig 3B and 3C ). These results indicated that the reduction in photosynthetic activity in leaves induced by OsWOX4 knockdown may be attributed to decreases in the accumulation of the chlorophyll-protein complex in photosystem II.

By contrast, lack of OsWOX4 resulted in a slight increase in vacuolation in the cells of the leaf primordia in the apical region of the P3 layer, and a little of the same effect was observed in the basal part of the P1 layer ( Fig 9L and 9N ). This suggests that OsWOX4 affects the cell cycle progression in the leaf primordia in a region-dependent manner. The cells of the leaf primordia are also degraded in barley, as reported by Reyes et al. (2013) [ 29 ]. The function of OsWOX4 seems to be related to those of barley BARLEY WUSCHEL (BWF) and Arabidopsis WUSCHEL (WUS) genes [ 39, 40, 44 ]. Our observation that the expression of OsARF11/BWF increases following the downregulation of OsWOX4 ( Fig 3D and 3G ) suggests that OsWOX4 functions downstream of the ARF family. Moreover, OsWOX4 regulates the expression of the orthologs of ARF family genes in the vascular bundle developmental stage and the basal part of P1 layer during leaf development in rice [ 24 ]. Further studies will be needed to reveal whether this mechanism also functions in barley.
