The V3 loop of the third variable region of HIV-1 gp120 has been considered as a dominant target for eliciting protective B-cell responses during HIV infection. A DNA vaccine based on the V3 loop of clade B strain (HIVac/B) was evaluated for its ability to elicit protective immune responses in rabbits. Immune responses were measured by an in vitro V3 loop-directed assay (V3 loop-specific ELISPOT) and by the detection of IgG reactive with the V3 loop in ELISA. Four immunizations with the DNA vaccine HIVac/B given with electroporation were effective in eliciting cellular and humoral immune responses, as compared with a control group immunized with the vector plasmid only. In this study, rabbits were immunized with the HIVac/B and treated by electroporation, and the results show that the cellular responses could be significantly increased by electroporation. More importantly, the antibody responses were induced by immunization with HIVac/B and could be boosted significantly by electroporation. The production of anti-V3 loop antibodies in rabbits was detectable after the first immunization with HIVac/B, and a booster was required. Following the second immunization, antibody levels in serum increased from 128 to 3605, and antibody levels in mucosal secretions increased from 127 to 8376. Controversy has erupted after a controversial advertising campaign to promote a park run.

Organisers of the Belfast City Marathon are under fire for promoting a park run, despite the death of one participant in recent weeks.

It emerged yesterday that a man, who is thought to have died after taking part in a nearby parkrun in Greencastle, County Tyrone, had a link to a marathon running club.

Rural communities in north and west Tyrone have been angered by the marathon organisation’s promotion of the run through social media and at a race morning event on Sunday.

The runner’s family said they were shocked at the lack of response after the accident and received no apology for the late promotion.

And members of Greencastle parish council are now demanding answers from the organisers.

The death of the Greencastle man is being investigated by police, but marathon organisers have confirmed that he was a member of the club.

When contacted by the Irish Independent, marathon organisers in Belfast insisted that the event is not associated with the Tyrone club.

However, organisers from the Greencastle club have questioned whether the parkrun was promoted at their event.

Members of the Tyrone club told the Irish Independent yesterday that they have written to the council demanding an explanation over the promotion.

They claimed that the two parks were visited by runners on the same day and they received “little if any” promotion.

This is despite the organisers of the parkrun promoting it on social media last week.

Organiser of the event, Diarmuid Cregg, who runs with the Tyrone club, said he was “flabbergasted” that organisers had promoted the event without telling him.

“The organisers are supposed to be helping their communities and providing a great service, not trying to steal the headlines,” he added
