Nitric oxide (NO) is an important cell signaling molecule and its biological function is mainly related to the isoforms of NO synthase (NOS), endothelial (e) and neuronal (n) NOS. Polymorphisms of NOS genes may be involved in the occurrence and development of tumors. In this study, we examined the genetic susceptibility to esophageal and gastric cancers (E&G cancers) in Chinese. Two polymorphisms of eNOS-786C>T and Glu298Asp were selected to analyze the association in a case-control study of 375 E&G cancer patients and 470 healthy controls. Data were statistically analyzed by SPSS 13.0 software. Our results showed that, compared with the wild homozygous 624C/C, the frequency of 624C/T in esophageal cancer was significantly higher (P=0.021), and the frequency of 624C/T in gastric cancer was significantly lower (P=0.008). The frequency of the 624C allele in esophageal cancer was significantly higher than that of the controls (P=0.031). No differences in frequencies of the NOS Glu298Asp variant were observed.
